Abstract<正>超低温保藏技术已被广泛使用于藻类的保藏。我们选择常见的水华蓝藻-铜绿微囊藻FACHB-905,FACHB-942和PCC7806作为实验材料,用二步法实现超低温保藏:使用细胞内冰冻保护剂(甲醇,DMSO和甘油)联用细胞外保护剂polyvinylpyrrolidone(PVP),每一种保护剂设定了4个浓度梯度(5%,10%,15%,22.5%)(V/V),降温速率设定了两种(-1℃min~(-1),-0.5℃min~(-1)),第一步降温达到-30℃,-40℃或-80℃,解冻后使用流式细胞仪以FDA染色
