AbstractThe present invention discloses a preparation method of sterile transgenic fish, and said method includes the following steps: using carp actin gene promoter as promotor, using antisense 296 bp carp salmon type gonadotropin-releasing hormone gene complementary DNA fragment [including sGnRH+peptide and gonadotropin-releasing hormone related peptide coding zone and 3' non-coding zone sequence of 64 bp] as target gene, and using grass carp growth hormone gene 3' flanking sequence as termination sequence to construct salmon type gonadotropin-releasing hormone antisense RNA expression vector, and adopting microinjection method to introduce the recombinant DNA fragment into carp fertilized egg, and utilizing PCR reaction and radioimmunodetection to screen and obtain the invented sterile transgenic fish.
